Seminar: RNA in Focus – From Extraction and Quantification to Protein Expression

The seminar will provide a comprehensive overview of RNA workflows and their integration with protein level gene expression analysis.

The first part will focus on the key steps in RNA analysis, including RNA extraction, the benefits of automation, quality control, and reverse transcription. Greater emphasis will be placed on qPCR and the fundamentals of qPCR data analysis.

The second part will address the relationship between mRNA abundance and final protein levels. Because gene expression is regulated at multiple post-transcriptional, translational, and post-translational levels, elevated transcript abundance does not necessarily result in increased protein expression. In this context, HiBiT technology will be introduced as a sensitive tool for monitoring total cellular protein levels, protein stability, and protein degradation. HiBiT application in RNA therapeutics will also be discussed, particularly for evaluating the impact of mRNA modifications on subsequent translation.

The seminar will further cover RNA interference (RNAi) as both a regulatory mechanism of gene expression and a valuable tool in the development of RNA based therapeutics. The mechanism of RNAi action will be presented together with siRNA pooling strategies aimed at improving silencing efficiency while minimizing off target effects.

Finally, we will show how the effects of RNA therapeutics can be assessed using cell based assays focused on proliferation, viability, and apoptosis.
